got a 1/4 inch NPTF port on a manifold block and need to run a JIC line off it, this fitting do the trick?

That's exactly what this adapter is built for. One end is a 1/4-18 Female NPTF, so it threads right into that port. The other end is a 1/2-20 Male JIC with a 37-degree flare seat, which connects to your JIC hose or tube assembly. Rated to 6,000 psi, so it holds up well in most hydraulic circuits without issue.

got a hose end that’s male JIC 37 degree flare, size 1/2-20. will this screw into it and give me a 1/4-18 female pipe port?

With a #5 MJIC (male JIC 37 degree flare) on one side and a #4 female NPTF (1/4-18) on the other, this fitting is made to adapt from a 1/2-20 JIC flare connection over to a 1/4-18 female pipe thread port. The JIC end follows SAE J514 and the NPTF end follows SAE J476.
