fitting's got a 1/4 inch barb on one end, does the other side thread straight into a standard 1/8 NPT port?

That 1/8-27 male NPTF thread on the pipe end is built to SAE J476 spec, so it'll seat properly in any standard 1/8 NPT female port. The NPTF thread form is a dry-seal design, meaning it relies on thread interference to hold pressure rather than just a sealant, though thread sealant is still commonly applied in practice. The barb end accepts 1/4 inch OD hose, so as long as your hose matches that OD and your port is 1/8 NPT female, the connection lines up on both ends.
