Got a 1/4-inch tube line I'm trying to branch off of, will this tee work for that setup?

A 0.25-inch tube O.D. is exactly what this fitting is sized for, so yes, it'll mate up correctly. All three ends are male SAE 45-degree flare, meaning you'll need female flare connections on whatever lines you're joining into.
