Got a 3/8 NPT female port that needs to connect to 1/4 NPT male fitting - will this work backwards or do I need something else?

You'll need a different adapter for that setup. Both ends of this particular fitting are male NPT threads - one end is 3/8-18 male and the other is 1/4-18 male. Since you have a female port and need to connect to a male fitting, you'd want an adapter with at least one female end to accept your male fitting.
